I figured it had to do with mirrors in the second telescope and vampires don't show up in mirrors, but found it kind of dumb and contrived. Meh.
 
Personally I think there's a technical error in how it was drawn that makes the joke a lot less obvious than it would otherwise be. With all the other attention to detail I'm surprised Phillips overlooked the viewing angle for the equipment suggested.

Compact telephoto lenses are designed that way, so it didn't hinder me.

While a reflector usually has a side viewing position that is because that's more convenient for the user. It's quite possible to have a reflector where the light comes out the end rather than the side. The telescope still needs to point at the object it's operator wishes to observe.

But I still don’t understand the need to make it about vampires.
There are other supernatural creatures which reportedly don't show up in mirrors, but the joke would get even more obscure very quickly.

Duh. I’m such an idiot. I was thinking of the aperture. With a large Schmidt Cassegrain, you’re looking at a smaller field of view and magnified so much more. So you wouldn’t see but a small part of the object and thus not recognize what it was.

That's dumb. Scientologists don't go door to door asking people to talk about scientology. You're thinking of Jehovah's Witnesses or some other strain of evangelical Christians. Scientologists prefer accosting people in downtown areas offering a free film or free personality test.

And they never mention Scientology, either, in my experience. That would scare off many potential suckers converts.
